biomacromolecules remains a challenge. In recent years, combinatorial library strategy
has been utilized to synthesize lipid-like materials for therapeutics delivery,
including proteins and nucleic acids. The development of combinatorial lipids with
various tails largely enriched the availability of therapeutics delivery. In this
thesis, I generated a combinatorial library (18) of lipids containing fluorinated tails.
We investigated the potential applications of these novel fluorinated lipids for
intracellular mRNA and protein delivery. GFP-Cre protein and firefly luciferase mRNA
were chosen as model cargos for the delivery. We successfully identified several
fluorinated lipids showing higher delivery efficiency than commercial reagent
Lipofectamine 2000. These fluorinated lipids also showed excellent
cytocompatibility.read less
